Profluent seeks a Scientist II to lead development of non-viral delivery methods for large gene insertion using recombinase-based approaches.   • Strong hands-on experience in molecular cloning, nucleic acid construct design, and mammalian cell engineering
  • Deep understanding of nucleic acid delivery barriers in mammalian cells and therapeutic applications
  • Extensive experience with NGS-based assays for evaluating genome editing outcomes
  • Demonstrated ability to develop mechanistic cell-based assays and troubleshoot complex biological systems
  • Proven success leading multifaceted research projects independently with strong communication skills

What you'll be doing

  • Lead development, evaluation, and benchmarking of non-viral donor template delivery methods for recombinase-based large gene insertion
  • Design and execute hypothesis-driven experiments to identify and overcome barriers to donor template stability, trafficking, and nuclear availability
  • Develop quantitative assays connecting donor template delivery measurements to insertion efficiency and fidelity
  • Analyze data to inform go/no-go decisions and present conclusions to cross-functional stakeholders
  • Validate delivery approaches in therapeutically relevant cell-based and in vivo systems with translational teams
